Questions How to make seeds from autos

I've never done it, but I'll outline the theory below.  Keep in mind, I am not an expert, I just have some detailed knowledge. YMMV. Maybe @Monkeydo can chime in and offer any corrections.

Start your donor individuals 2 to 2 1/2 weeks before you start your recipients. If you want only autos, then both donor and recipients must be autos since it is recessive, else your crop will only be carrying one auto gene and not be an auto.  Isolate the donor and start spraying with STS at 4 weeks; spray once a day every 5 days only at the elbows. The spray will likely damage the plants being reversed. Most people will get in at least 3 sprays, but this is strain-dependent. Given the odds of reversal, you are more likely to succeed if you reverse several small donors instead of one big one.  Success and reaction also are very strain-dependent, partly due to plant structure and bud habit. Introduce the recipients into the donor's tent when pistils are fully emerged, not when they first emerge. Shake the donor over the recipient once a day for a couple of days. Watch for seed development.  Allow recipients longer time to fully develop seeds. DO NOT smoke any of the sprayed bud. Plain water will inactivate the pollen. Reversed plants typically produce only a tiny fraction of that of a standard male plant. 

Note, even world-class breeders and seed-makers have frequent failures with reversal. Even the same strains treated the same manner will have differential success for unknown reasons. It is part of the process. You have to work the odds.

Well done Sparky. Only a couple of details to put in. Your timing is correct. Start the pollen recipient 2 weeks after the donor plant but because of the variability in strains and how they take to the STS, I spray 6 times with 5 days between sprayings and get very good reversals like this. Some strains reverse completely at one spray, some only partially reverse after 6 sprays but you always get enough of a reversal with 6 treatments to make seeds. To increase the seed yield I keep the pollen donor in a small pot, usually a solo cup, and simply pick her up and physically touch the recipient plants pistils with the reversed flowers for sure thing pollination. After 3-4 days of this you are done with the reversed plant and it can be tossed. Removing the pollen donor quickly after pollination ensures all the seeds will be a similar maturity at harvest. The seeds need a minimum of 6 weeks on the plant to mature and longer is better. If you want the classic tiger striped seeds it definitely takes a little longer. Be aware that one small plant can make many hundreds or even thousands of seeds. Good luck and please come back and show us how you did. Could I take one plant spray half with sts and just allow it to pollinate the other half it’s self. Or do I need the separate plant in order to do it?

@bobblehead yes it is possible but definitely not an ideal situation. Fully reversing a bud takes about a month and the unsprayed branches would be starting to flower while that branch is reversing. By the time that branch starts making pollen you’ll be 4-6 weeks into flower. That’s not going to leave a lot of time for the seeds to mature before harvest. It is done with varying degrees of success but it’s not ideal at all. The resulting seeds might be immature or duds. I just planted my second Mephisto beans yesterday for that seed making process I’m currently doing. It might seem wasteful to you using another seed, but if you are successful you’ll end up with hundreds or even thousands of seeds.
